摘要
目的 改进体外培养牛眼小梁细胞的方法。方法 分别采用常规和改进的方法对牛眼小梁细胞进行体外原代及传代培养 ,比较两者在组织成活率、细胞贴壁率、生长曲线上的差异 ,并做组织学特异染色鉴定 ,应用光学显微镜、电子透射显微镜对原代及传代培养的细胞进行形态学及生长特性的观察。结果 牛眼小梁细胞培养成功 ,两种方法对培养无明显影响。结论 应用加有小牛血清的培养液可在不影响培养效果前提下明显降低实验成本 。
Objective To improve the method of culturing bovin trabecular meshwork cells(BTMC) in vitro.Methods The BTMC were cultured by general and improving method, comparing with surviving efficiency, attachment efficiency and growth curve, and cells were identified by immunohistochemisty method (NSE and Ⅷ factor related antigen). The growing characteristics and morphological feature of cultured primary and passaged cells were observed by inverted microscope and electron-microscope.Results The cells were cultured successfully. The live rate of trabecular meshwork cells was 80%～100%. Between them there is no significant difference for culturing.Conclusion It could obviously reduce experimental benefit-cost ratio to culture with newly born bovin serum. This study indicated that selective collecting tissue of trabecular meshwork is a precondition for successfully culturing BTMC.
