摘要
目的:观察铸造钴铬合金含银抗菌涂层对小鼠成纤维细胞(L929)增殖及凋亡的影响。方法:应用等离子喷涂技术制备含银抗菌涂层钴铬合金试件10个,制作不含涂层的钴铬合金试件10个作为对照,用2组试件的浸提液培养L929细胞24、48和72 h后,利用MTT法检测细胞增殖情况;通过免疫细胞化学染色观察用浸提液原液处理48 h后L929细胞中Bcl-2和Bax蛋白的表达情况。结果:经含银抗菌涂层钴铬合金试件浸提液培养后,L929细胞增殖状态与对照比较差异无统计学意义(P〉0.05),浸提液对L929的细胞毒级为0-1级;细胞内Bcl-2和Bax蛋白表达水平与对照比较,差异也无统计学意义(P〉0.05)。结论:钴铬合金含银抗菌涂层具有良好的细胞相容性。
Aim:To evaluate the influence of casting cobalt chromium alloy Ag-containing coating on L929 cell proliferation and apoptosis. Methods:Using the technique of plasma spraying,10 pieces of casting cobalt chromium alloy Agcontaining coating specimens as experimental group and 10 pieces of cobalt chromium alloy specimens as control group were used to prepare the leaching liquor. The MTT and immunohistochemistry method was adopted to detect proliferation and the expression of Bcl-2 and Bax proteins of the L929 cells treated by the leaching liquor. Results:The cytotoxic level of the leach liquor of casting cobalt chromium alloy Ag-containing coating pieces was Level 0- 1. There was no statistical differences in the proliferation of L929 cells or the expressions of Bcl-2 and Bax protein for 2 kinds of leaching liquor( P 0.05). Conclusion:The cell compatibility of Ag-containing on casting cobalt chromium alloy is well.
