摘要
目的:探讨不同状态下大鼠星形胶质细胞(AS)条件培养液对脑微血管内皮细胞(BMEC)活性和血脑屏障特征性酶的影响,以及通络救脑注射液对此过程的干预作用。方法:收集正常、激活、激活合并通络救脑注射液处理、损伤、损伤合并通络救脑注射液处理5种不同状态的星形胶质细胞条件培养液,分别作用于拟缺血损伤的脑微血管内皮细胞,采用MTT法检测细胞活性,微量酶标仪法和上机比色法检测碱性磷酸酶(AKP)、γ-谷氨酰胺转肽酶(γ-GT)的活性。结果:正常星形胶质细胞条件培养液能明显提高受损内皮细胞活性,诱导AKP、γ-GT活性增强,激活的星形胶质细胞条件培养液上述作用下降,而损伤后的星胶条件培养液对受损内皮细胞几乎无作用。通络救脑注射液作用于激活和损伤星形胶质细胞后,其条件培养液提高内皮细胞活性和AKP、γ-GT活性的作用显著增强。结论:正常AS的分泌产物对提高BMEC的活性和维持血脑屏障特性具有重要意义,AS受损后,该作用减弱或消失,通络救脑注射液可显著提高AS对受损BMEC活性和功能的保护作用,这可能是该药物发挥脑缺血性损伤后血脑屏障稳定作用的内在机制之一。
This study was aimed to observe the effect of conditioned media from astrocytes on the rat brain microvascular endothelial cells (BMEC), including the activity of BMEC, AKP and γ-GT, as well as to investigate the influence of Tong Luo Jiu Nao (TLJN) injection. The conditioned media from normal, activated, activated and TLJN administrated, injured, injured and TUN administrated astrocytes were collected. Then 5 conditioned media were added into injured BMEC cultures respectively. The effect was determined through the measurement of MTT and AKP, T-GT Kit. The results showed that conditioned medium from normal astrocytes significantly increased the activity of BMEC, AKP and γ-GT. The conditioned medium from activated astrocytes showed similar function, but it was weaker than normal astrocytes. The injured astrocytes conditioned medium did not show any function on injured BMEC. In the groups treated with TUN, BMEC activity and the expression of AKP, γ-GT was higher than groups without treatment. It was concluded that the secretion of normal astrocytes can highly increase the activity of BMEC, AKP and γ-GT. As the astrocytes were activated and injured, this function would be decreased or disappeared. TLJN can observably improve the function of astrocyte on the activity of BMEC, AKP and γ-GT, which may be the mechanism of TLJN on the protection of blood-brain barrier.
