POP患者子宫骶韧带组织中胶原纤维、弹力纤维形态学变化及HOXA11、ER-α和ER-β的表达及其意义 被引量:4

摘要 目的:探讨子宫骶韧带局部组织中同源盒基因A11(HOXA11)、雌激素受体α(ER-α)、雌激素受体β(ER-β)的表达及胶原纤维、弹力纤维形态的改变与盆腔器官脱垂(POP)发生的关系。方法:采用免疫组织化学PV法检测24例(POP及非POP各12例,分别为研究组和对照组)子宫骶韧带组织中HOXA11、ER-α、ER-β的表达,应用间苯二酚品红法和丽春红酸性品红法分别对弹性纤维和胶原纤维进行染色。结果:①HE染色显示研究组子宫骶韧带组织有胞浆水肿、空泡变性及淀粉样变等不同程度的变性。②与对照组相比,研究组胶原纤维染色较浅,弹力纤维弯曲度和密度减低,纤维碎裂、崩解或聚集成团块状,排列相对不规则。③研究组HOXA11、ER-α和ER-β表达均减弱,与对照组相比,差异有统计学意义(P<0.05)。结论:POP患者子宫骶韧带组织发生变性,其弹力纤维的形态遭到破坏,胶原纤维的含量减少。HOXA11、ER-α和ER-β在POP患者子宫骶韧带组织中的表达均降低,提示均参与POP的发生。 Objective:To investigate the expression of HOXA11,estrogen receptor alpha,beta and morphologic changes of collagen and elastic fibres in uterosacral ligament of patients with pelvic organ prolapse(POP),to explore the relationship of above parameters with POP.Methods:The expression of HOXA11,estrogen receptor alpha and beta were detected by immunohistochemistry(PV method).Collagen fibres and elastic fibres were stained by ponceau acid fuchsin and resorcin fuchsin methods.Results:①Endochylema hydrosarca,vacuolar degeneration and amyloid degeneration were found in uterosacral ligament of POP group under HE stain.② More pale color collagen fibres,hypocurvature,hypodensity,broken or mess elastic fibers arranged more irregularly in POP group than that in control group.③ The expressions of HOXA11,estrogen receptor alpha and beta were significant lower in POP group than those in control group(P0.05).Conclusions:The uterosacral ligament of POP patients has degenerative changes,the morphology of elastic fiber is damaged,the quantity of collagen fibres is reduced.The expressions of HOXA11,estrogen receptor alpha and beta are all reduced in uterosacral ligment of POP group,which hints these factors may involve the pathogenesis of POP.
