茶树不同组织体细胞胚、不定芽分化的研究 被引量:18

摘要 根据茶树子叶柄、叶柄断面离腋芽的距离,将外植体依次分为C_1、C_2、C_3和L_1、L_2、L_3.通过上述外植体和不同培养基的研究结果表明:子叶柄外植体在MT_1和MT_3培养基上培养,其不定芽及总分化率是C_1>C_2>C_3;体细胞胚的分化率是C_3>C_2>C_1.叶柄外值体只有L_1在MT_3培养基上培养分化了芽,MT_1培养基的胚性愈伤组织诱导率比MT_3培养基高,其体细胞胚、不定芽的分化率是L_1>L_2、L_3.经挑选的胚性愈伤组织在MT_1和MT_2培养基上交叉培养,保持分化能力在15个月以上.体细胞胚、不定芽都能在MT_4培养基上增殖. The explants of tea plant were classified into 6 types i. e. C1、C2、 C3and L1、 L2、L3,according to the distances from cut of cotyledon petiole and leafstalk to axillary bud. The effect of the explant types on differentiation rates of somatic embryo and adventitious bud was studied. Adventitious bud differentiation rate and total differentiation rate were C1>C2>C3, but somatic embryo differentiation rate was C3>C2>C1, when the explants were cultured on medium MT1 and MT3. Only explant L1 which were cultured on medium MT3 differentiated bud in the leafstalks culture. Number of the leafstalk explants producing embryogenic callus on medium MT1 was more than that on medium MT3. The differentiation rate of leafstalk explants was L1>L2、 L3.When embryogenic callus was carefully selected and alternately subcultured on medium MT1 and MT2, it could retain plant regeneration capacity for 15 months or more. Adventitious bud and somatic embryo could proliferate on medium MT4.
