比色法测定野马追糖浆总黄酮的含量

摘要 目的:建立野马追糖浆总黄酮的含量测定方法并确定其限度。方法:以芦丁为对照品,亚硝酸钠-硝酸铝显色后在510nm波长处测定吸收度的比色法测定总黄酮含量,并对实际产品含量进行考察。结果:在浓度0.0108~0.0540mg·mL-1范围内线性良好,r=0.9998;平均加样回收率为99.68%,RSD为1.13%;野马追糖浆实际产品总黄酮含量在2.59~3.92mg·mL-1之间。结论:本研究提出的含量测定方法准确稳定、简便易行,经方法学验证符合要求。总黄酮含量限度确定为以芦丁(C27H30O16)计不得少于2.0mg·mL-1。 Objective To establish the assay method and determine the limit of the total flavonoids in Yemazhui Syrup.Method The total flavonoids content of Yemazhui Syrup was determined by spectrophotometry detecting absorbances at 510 nm with aluminum nitrate as the colour- developing agent and Rutin as the reference substance. Results The total flavonoids had a good linear relationship within the range of 0.0108- 0.054mg·mL^-1 (r= 0.9998);The mean recovery was 99.68%,and the relative standard deviation (RSD) was 1.13%;The contents of total flavonoids varied from 2.59-3.92 mg· mL^-l. Conclusion The assay methods established in this study was accurate,stable and convenient, complying with the requirements validated by methodology.The assay limit of total flavonoids was set as not less than2.0 mg·mL^-1 caculating based on Rutin.
