摘要
目的探讨核因子κB(NFκB)抑制剂吡咯烷二硫氨基甲酸(PDTC)对肺癌裸鼠移植瘤生长及血管生成的影响。方法人肺癌A549细胞接种于裸鼠皮下,建立肺癌裸鼠移植瘤模型,分为对照组和PDTC组,比较移植瘤重量,计算抑瘤率;RTPCR检测移植瘤环氧化酶2(COX2)基因表达,免疫组织化学法检测血管内皮生长因子(VEGF)和FⅧ蛋白表达,并计数微血管密度(MVD)。结果PDTC组移植瘤重量显著低于对照组(P<0.01),抑瘤率为41.03%;COX2、VEGF表达及MVD计数均显著低于对照组(P<0.01,P<0.01,P<0.05)。结论PDTC可抑制肺癌裸鼠移植瘤的生长,该作用可能与其抑制COX2、VEGF表达,从而抑制血管生成有关。
Objective To study the effect of pyrrolidine dithiocarbamate (PDTC), a potent inhibitor of nuclear factor κB (NF-κB) activation, on the growth and angiogenesis of human lung cancer xenografts in nude mice. Methods A549 cells were incubated and were inoculated subcutaneously in athymic nude mice to establish xenograft models.The mice were divided into control group and PDTC group. The inhibitory rate was calculated according to the weights of xenografts. Cyclooxygenase 2 (COX-2) mRNA was measured by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) analysis. The proteins expressions of vascular endothelial growth factor (VEGF) and FⅧ were assessed by immunohistochemical method, and MVD was counted according to FⅧ staining. Results The weight of xenografts was significantly lower in PDTC group than that in control group (P< 0.01) and the inhibitory rate was 41.03%. The expressions of COX-2, VEGF and MVD count of xenografts were significantly decreased in PDTC group than that in control group (P< 0.01,P< 0.01,P< 0.05). Conclusion PDTC can inhibit the growth of human lung cancer xenografts in nude mice, which may be due to the downregulation of COX-2 and VEGF thus inhibit tumor angiogenesis.
