摘要
通过对几种培养基比较试验,筛选出WPM是早熟油桃胚培养的最适培养基,其最高萌发率为95.56%;不同熟期成熟的早熟油桃品种对低温处理的要求时间不同,愈是早熟的品种对低温要求时间愈长,晚熟品种相对较短;1mg/L 6-BA+0.2mg/LIBA+0.1 mg/L GA。能有效破除早熟油桃的休眠,而且幼苗生长良好。早熟油桃胚培养培养基的pH值要求范围为5.7-6.2,其中愈是早熟对培养基pH值的要求相对较高;对试管苗的移栽采用白天口杯罩住,夜间揭开,持续5-7 d,3 d多菌灵灌根1次,15 d后发现腐烂子叶及时清除,能有效提高试管苗的成活率。
Different media were used to rescue embryos of early ripening nectarines (Primus persica var. nectarina Maxim). Results showed that WPM was the best medium for rescuing embryos. The germination percentage was the highest, reached 95.56% when WPM was used as the culture medium. Different cultivars required different time of low temperature treatment to germinate, the earlier the cultivars ripened, the longer the time they needed. The combination of 1 mg/L 6-BA + 0.2 mg/ L IBA + 0.1 mg/L GA3 broke embryo dormancy of nectarines effectively, and seedlings from treated embryos grew well. pH in medium was from 5.7 to 6.2, and the early ripening cultivars needed relatively higher pH in medium for embryo rescue. Plastic cups were used to cover the seedlings from embryo rescue in the daytime and cups were removed at night for 5 to 7 d for training the seedlings. Fungicide was sprayed into the soil every 3 d, and 15 d later, the rotten cotyledons were removed in time. With the methods above, the survival percentage of the rescued seedlings was increased.
