摘要 由淀粉通过葡萄糖异构化生产高果糖糖浆,生产上受到原料的限制。果糖另一种来源是菊粉(Inulin)。菊粉是以β-2,1果糖苷键连接的一种多聚果糖,其末端含有一个蔗糖残基,它作为贮存性多糖大量存在于菊芋(Helianthus tuberosus)、菊巨(chicoryintybus)、大丽花(Dahlia pinnata)等多种植物中,至今尚未得到很好利用。菊粉可通过化学法或酶法水解生成果糖。 A high inulinase producing fungus Penicillium SP. 91-4 was isolated from the soil. The effect of nutrients and cultural conditions on inulinase producing strain 91-4 was analysed.The optimum culture conditions were as follows:initial pH6.0, temperature 30℃, and cultivation time 72 hr. The highest inulinase activity (50.0 units/ml) was obtained under the optimum conditions. The optimal pH and temperature for inulinase reaction were pH 5.6 and 45℃. Inulinase was stable within the pH range 5.0-7.0, and below the temperature 45℃. The inulinase activity was inhibited strongly by PCMB (P-chloromercuri benzonic acid) and activated by Mn2+. The hydrolysis of Jerusalem artichoke extracted juice by crude extracellular enzyme was investigated.
