摘要
用印迹法和半粒法对胡麻脂肪酸变异系E1747与内亚3号正、反交后代种子无胚端亚麻酸含量进行了定性鉴定,并使入选的低亚麻酸含量种子的胚芽端再生幼苗。实验结果表明,采用印迹法定性鉴定正、反交后代种子,筛选无胚端油斑染色后颜色明显比亲本内亚3号浅的,即亚麻酸含量低的材料,再采用半粒法使其胚芽端再生幼苗,这是进行低亚麻酸材料筛选、保存与繁育的一种可靠方法。半粒法再生幼苗过程如下:无菌条件下将种子横切成两半→胚芽端接种到有无菌湿滤纸的培养皿内→20℃、光照培养16h和15℃、黑暗培养8h→将幼苗移至小营养钵中→在室内阴暗处缓苗1d→移至室温光照条件下培养→幼苗至5cm高度时移至大营养钵中。
This paper evaluated qualitatively the ALA content of non-germ-ends of seeds from reciprocal cross between the fatty acid variant line E_(1747) and Neiya No.3 of linseed through the spot-test and the half-seed method, as well as making germ-ends of selected low-ALA seeds regenerating seedlings. The result showed that we evaluated qualitatively reciprocal cross offspring seeds through spot-test, and we screened the materials that the oil spot color of non-germ-ends through staining lighter than parent of Neiya No.3. The ALA content of materials was low. We cultured these germ-ends of materials and made them regenerate seedlings. The method that we screened, preservated and bred low-ALA materials was reliable. The procedure of regenerative seedlings by the half-seed method as follows: traversing the seed into halves under aseptic condition→inoculating the germ-end on the sterilized-wet filter in culture dish→culturing for 16h under light at 20℃ and for 8h under darkness at 15℃→transplanting the seedling into the smaller nutritive cube→training the seedling for one day in shady condition→culturing the seedling under room temperature and light condition→transplanting the seedling into the bigger nutritive cube, when it grew to the height of 5cm.
