摘要
根据西瓜花叶病毒2号(WMV 2)的外壳蛋白基因序列设计、合成引物,以感病组织和健康组织总RNA为模板,含WMV 2外壳蛋白基因的质粒DNA为阳性对照,进行cDNA合成和PCR扩增。结果从感病组织中扩增出与预期的382bp大小一致的目标片段,而健康组织无此扩增产物。对2002和2003年采集的98份黄瓜病毒病样本进行了同样的RT PCR检测,结果表明98份材料中77.55%检测到WMV 2。
A pair of primers were designed and synthesized based on the RNA sequences of WMV-2 coat protein, and RT-PCR was applied to total RNA detection which was extracted from watermelon mosaic virus-infected and healthy cucumber leaves. A DNA fragment of about 382 bp was amplified from the infected samples, but not from the healthy samples. A total of 98 cucumber leaves suffered from viruses and collected from several regions of Tianjin in 2002 and 2003 were detected. The results show that the incidence rate of watermelon mosaic viruses-2 (WMV-2) was 77.55%.
