摘要
采用RAPD技术对66份荔枝古树(其中福州市64份,广东开平市2份)资源进行遗传多样性分析。结果表明,16个随机引物共产生232条RAPD带,其中216条为多态性带,占总带数的93.10%,说明66份荔枝古树资源有丰富的多样性。根据扩增条带多态性,利用UPGMA构建了荔枝古树的亲缘关系树状图,遗传距离为0.000~1.000。当D=0.909时,66份荔枝古树资源可以分为3组;当D=0.746时,分为9组;当D=0.578时,可归为18组,同一地方的荔枝古树基本可以划分在一起,但也有个别划分在不同组,亲缘关系较远,表明福州市区荔枝古树群体存在一定的遗传变异。该研究为福州市区荔枝古树特异基因资源的保护与进一步挖掘利用提供了科学依据。
Genetic diversity of 66 accessions of ancient litchi trees was evaluated by using RAPD (Random amplified polymorphic DNA) . The results showed that 232 RAPD bands were amplified by 16 random primers, among which 216 were polymorphic, 93.10% of the total bands, suggesting that 66 accessions of ancient litchi trees are abundant in genetic diversity.
