复合抗氧化剂对α-鹅膏肽毒素促 V79细胞凋亡的干预研究

摘要 目的研究α-鹅膏毒素对V79细胞的促凋亡作用,了解复合抗氧化剂维生素C(VC)与维生素E(VE)对α-鹅膏毒肽促V79细胞凋亡的干预作用。方法体外培养V79细胞,除对照组外,其余各组按终浓度5μg/mLα-鹅膏毒素染毒,早期干预组同时按终浓度100μmol/L加入VC和VE;后期干预组染毒24 h后加入同浓度VC和VE。检测细胞存活率、细胞线粒体膜电位,以及细胞MDA(丙二醛)及SOD(超氧化物歧化酶)含量并比较。结果与对照组相比,各组细胞存活率均降低,其中染毒组最低。细胞线粒体膜电位检测结果中,染毒组正常细胞数最少,早期干预组数接近对照组,而后期干预组数较少;染毒组细胞凋亡率明显高于对照组,早期干预组低于染毒组和阳性对照组。两个干预组MDA含量均低于染毒组,SOD含量均高于染毒组和对照组。结论α-鹅膏肽毒素可引起V79细胞凋亡,VC和VE的抗氧化组合可干预其致细胞凋亡作用;与后期干预相比,早期干预效果更加明显。 Objective To study the pro-apoptotic effect ofα-amanita toxin on V79 cells and to understand the intervention effect of compound antioxidants vitamin C(VC)and vitamin E(VE)onα-amanita toxin in promoting V79 cell apoptosis.Methods V79 cells were cultured in vitro.Except the control group,the other groups were exposed toα-amanita toxin at a final concentration of 5μg/mL.In the early intervention group,VC and VE were added at a final concentration of 100μmol/L at the same time.The late intervention group was also added with the same concentration of VC and VE after 24 hours.Cell viability,cell mitochondrial membrane potential,and cellular MDA(malondialdehyde)and SOD(superoxide dismutase)contents were detected and compared.Results Comparing with the control group,the cell viability in each group decreased and the exposure group was the lowest one.In the results of mitochondrial membrane potential,the number of normal cells of the exposed group was the least one,the number of early intervention group was close to the control group,and the number of late intervention group was less.The apoptosis rate of the poisoned group was significantly higher than that of the control group,and that of the early intervention group was lower than that of the poisoned group and the positive control group.The contents of MDA of the two intervention groups were lower than that of the exposure group,and the contents of SOD were higher than that of the exposure group and the control group.Conclusionα-Amanita toxin can induce the apoptosis of V79 cells,and the antioxidant combination of VC and VE can interfere with its apoptosis-inducing effect.Comparing with the later intervention,the effect of early intervention is more obvious.
