两种微量热技术定量检测蛋白质与化合物相互作用的特色比较及相互验证

Comparison and mutual verification of two microthermal techniques for quantitative detection of protein-compound interactions

摘要 分子互作技术通过检测蛋白质和其他物质相互作用的特性,如亲和力常数等探究分子间相互作用的机制。其中,等温滴定微量热技术(isothermal titration calorimetry,ITC)和微量热泳动技术(micro scale thermophoresis,MST)在分子互作定量检测中广泛应用。本文以一种磷酸甘油酸激酶1(phosphoglycerate kinase 1,P1)和药物分子盐酸特拉唑嗪(terazosin hydrochloride,TZ)之间的相互作用检测为例,运用ITC和MST技术顺利获得实验结果并可相互验证,同时对两种检测技术的优势和不足进行了深入探讨,为不同条件下检测方法的选择提供了新的思路。 The technologies are used to research the mechanism of molecular interaction by detecting the characteristics of the interaction between protein and other substances,such as affinity constant.The isothermal titration calorimetry(ITC)and micro scale thermophoresis(MST)are widely used in the quantitative detection of the interaction between moleculars.In this article,the two techniques are used for the quantitative detection of the interaction between phosphoglycerate kinase(P1)and terazosin hydrochloride(TZ),and the experimental results are successfully obtained and mutually verified.Meanwhile,the pros and cons of the two detection techniques are discussed in depth,which provides new ideas for the selection of detection methods under different conditions.
