摘要
目的探讨鄂尔多斯地区蒙古族老年人群SLCO1B1和ApoE基因多态性分布情况,为该地区蒙古族人群基因遗传学、心脑血管疾病和阿尔茨海默病的预防、服用他汀类药物疗效和致肌病风险评估提供理论依据。方法纳入120名鄂尔多斯地区蒙古族老年健康体检居民作为研究对象,采用聚合酶链反应-荧光探针法检测其SLCO1B1和ApoE基因位点多态性。结果所有入组样本基因片段均扩增成功,共检测到7种SLCO1B1基因型,分别为*1a/*1a(5.00%)、*1a/*1b(38.33%)、*1a/*5(0.83%)、*1a/*15(10.83%)、*1b/*1b(32.50%)、*1b/*15(10.83%)、*15/*15(1.67%),未检测到*5/*5和*5/*15基因型。共检测到5种ApoE基因型,分别为ε2/ε3(5.00%)、ε2/ε4(0.83%)、ε3/ε3(80.00%)、ε3/ε4(12.50%)、ε4/ε4(1.67%)基因型,未检测到ε2/ε2基因型。结论在鄂尔多斯地区蒙古族老年人群中,SLCO1B1基因以*1a/*1b基因型和*1b等位基因最常见,ApoE基因以ε3/ε3基因型和ε3等位基因最常见。SLCO1B1基因以正常肌病风险的人群为主(75.83%),大部分人群服用他汀类降脂药物为正常肌病风险。ApoE基因以野生基因型人群为主(80.83%),大部分人群患心脑血管疾病、阿尔茨海默病为正常风险,服用他汀类药物降脂的疗效多数正常。
Objective To investigate the distribution of SLCO1B1 and Apo E gene polymorphisms in elderly Mongolian population in Ordos area,and to provide theoretical basis for genetic genetics,prevention of Cardio-cerebrovascular disease and Alzheimer’s disease,efficacy of statins and risk assessment of myopathy in elderly Mongolian population in Ordos area.Methods A total of 120 Mongolian elderly health checkup residents in the Ordos region were included as the research objects,and their SLCO1B1 and Apo E gene polymorphisms were detected by polymerase chain reactionfluorescence probe method.Results The gene fragments of all the included samples were successfully amplified,and a total of 7 SLCO1B1 genotypes were detected,namely*1a/*1a(5.00%),*1a/*1b(38.33%),*1a/*5(.83%),*1a/*15(10.83%),*1b/*1b(32.50%),*1b/*15(10.83%),*15/*15(1.67%),not detected*5/*5 And*5/*15 genotypes.A total of 5 Apo E genotypes were detected,namelyε2/ε3(5.00%),ε2/ε4(.83%),ε3/ε3(80.00%),ε3/ε4(12.50%),ε4/ε4(1.67%)Genotype,ε2/ε2 genotype was not detected.Conclusion In the Mongolian elderly population in Ordos,the SLCO1B1 gene is the most common with the*1a/*1b genotype and*1b allele,and the Apo E gene is the most common with theε3/ε3 genotype and theε3 allele.The SLCO1B1 gene is dominated by people with normal myopathy risk(75.83%),and most people taking statin lipid-lowering drugs are at normal myopathy risk.The Apo E gene is dominated by the wild genotype population(80.83%).The majority of the population suffers from cardiovascular and cerebrovascular diseases and Alzheimer’s disease as normal risks.The effect of taking statins for lowering lipids is mostly normal.
