rhBMP_2在去势大鼠中的异位诱导成骨效应

摘要 目的:通过观察rhBMP2在去势大鼠中的异位诱导成骨效应,探讨rhBMP2与绝经后骨质疏松的关系。方法:对双侧卵巢切除和单纯行开腹术大鼠双侧股部肌袋植入1.5mg和3mgrhBMP2,于植入后1周、2周和3周取材,行组织学检查,成骨量分析,碱性磷酸酶和钙含量测定。结果:组织学检查发现rhBMP2剂量越大,成骨多,成骨早。实验组较对照组诱导成骨活性增强,表现为成骨量,碱性磷酸酶和钙含量较对照组明显增多(P<0.05)。结论:在卵巢切除大鼠早期rhBMP2表现较高诱导成骨活性,可能与机体代偿有关。 Objective: To investigate the relation beween rhBMP-2 and postmeno-pausal osteoporosis by researching the ectopic inductive bone formation activity of rhBMP-2 in ovariectomied (OVX) rats. Methods: 1.5mg and 3mg rhBMP-2 were implanted into the thigh muscle pouches of bilateral ovariectomied (OVX) rats and sham-operation rats. The rats were sacrificed after day 7.14 and 21.then the induced ectopic bone formation was studied by histological analysis,the quantity of new bone,the ALP activity levels and calcium content measurement. Results: The larger the dosage of rhBMP-2 ,the more the quantity of new bone and the earlier the bone formed. The OVX group was better than the control in histological analysis ,the quantity of new bone ,the ALP activity levels and calcium content. Conclusion:In early stage,the OVX rats manifested higher ectopic bone activity which the control, that showed some relation between rhBMP-2 and postmenopausal osteoporosis.
