摘要
目的分析血红素加氧酶1(HMOX1)在泛癌中的表达及其在肿瘤预后中的作用。方法采用肿瘤免疫数据库TIMER2.0和GEPIA2工具分析HMOX1在各癌症中的表达,探讨不同癌症中HMOX1表达与癌症相关纤维细胞浸润水平的关系;采用GEPIA2数据库分析不同癌症中HMOX1表达与癌症总生存期和无病生存期的关系;采用STRING和GEPIA2数据库得到HMOX1的相关蛋白互作网络图。结果HMOX1在癌症中的发病机制与炎症反应、细胞黏附有关,大多数基因编码定位在质膜上,并发挥三磷酸鸟苷结合和蛋白结合的分子功能。HMOX1在多种癌症中高表达,并且与肿瘤患者预后存在相关性;HMOX1的表达与膀胱尿路上皮癌、结肠癌、头颈鳞状细胞癌、脑低级别胶质瘤、胰腺癌、胃癌、睾丸生殖细胞癌等癌症相关成纤维细胞浸润水平呈正相关。结论HMOX1的表达与多种癌症临床预后、免疫浸润等存在相关性,通过泛癌分析,有助于从临床肿瘤样本角度阐述HMOX1在肿瘤发生中的作用。
Objective To analyze the expression of heme oxygenase 1(HMOX1)in generalized carcinoma and its role in tumor prognosis.Methods TIMER2.0 and GEPIA2 were used to analyze the expression level of HMOX1 in various cancers,and to explore the potential relationship between the expression level of HMOX1 and the level of cancer-related fibrocyte infiltration in different types of cancer genome map.GEPIA2 database was used to analyze the relationship between HMOX1 expression and overall survival and disease-free survival in different cancers in the cancer genome map.The interaction network diagram of HMOX1 related proteins was obtained by using STRING and GEPIA2 database.Results The pathogenesis of HMOX1 in cancer was related to inflammatory response and cell adhesion,and most of the gene codes were localized on the plasma membrane and played the molecular functions of guanosine triphosphate binding and protein binding.HMOX1 was highly expressed in a variety of cancers,and was correlated with the prognosis of tumor patients.The expression of HMOX1 was positively correlated with level of fibroblastsinvasion in bladder cancer,colon cancer,head and neck squamous cell carcinoma,brain lower grade glioma,pancreatic cancer,stomach cancer and testicular cancer.Conclusion The expression of HMOX1 is correlated with the clinical prognosis and immune invasion of a variety of cancers,and pan-cancer analysis is helpful to elucidate the role of HMOX1 in tumorigenesis from the perspective of clinical tumor samples.
