赤大袋鼠馆气载需氧菌分离鉴定及致病性分析

摘要 本试验采用自然沉降法对南京红山森林动物园赤大袋鼠馆的运动场、缓冲道、饲养内舍、工作通道和饲料间5个区域一年四季的气载需氧菌浓度进行测定,通过16SrRNA基因和生理生化鉴定相结合的方法分析气载需氧菌的组成。通过动物致病性试验对所分离的细菌进行致病性分析,采用药敏试纸片法分析致病菌株的耐药情况。结果显示,赤大袋鼠馆各个区域一年四季气载需氧菌浓度与温度呈正相关。经生化鉴定及16SrRNA比对结果分析,共鉴定3大类细菌71株,其中葡萄球菌属共8种共37株,占分离菌的52.1%;肠杆菌科细菌9个属,10种共31株,占分离菌的43.7%;链球菌属细菌仅分离到2种3株,占分离菌的4.2%。71株菌中,从饲养内舍分离出44株,其他区域共分离到27株,分离率分别为62.0%和38.0%。经致病性分析,2株大肠埃希菌,1株弗氏志贺菌,2株金黄色葡萄球菌及1株巴黎链球菌,共计6株菌为致病菌。分离的6株致病菌均对克林霉素耐药,对环丙沙星、氟苯尼考、庆大霉素、恩诺沙星和磺胺甲基异恶唑敏感。 We used the natural sinking method to determine the concentration of airborne aerobic bacteria in five areas of the red kangaroo exhibition,viz. playground,buffer path,indoor room,management channel,and storeroom,at Nanjing Hongshan Forest Zoo throughout the year. The composition of airborne aerobic bacteria was analyzed by 16S rRNA gene analysis and physiological and biochemical identification. The pathogenicity of the isolated bacteria was analyzed by animal pathogenicity testing,and the drug resistance of the pathogenic bacteria was analyzed by antimicrobial susceptibility testing. The concentration of airborne aerobic bacteria was positively correlated with temperature throughout the year in all five areas of red kangaroo exhibition. The results of biochemical identification and 16S rRNA comparison showed that 71 strains of three major types of bacteria were identified, including 37 strains of Staphylococcus representing 8 species, accounting for 52% of the isolated bacteria; 31 strains of Enterobacteriaceae representing 9 genera and 10 species,and accounting for 44% of the isolated bacteria; and only 3 strains of Streptococcus representing 2 species and accounting for 4% of the isolated bacteria. Among the 71 strains,44 strains were isolated from the indoor room,and 27 strains were isolated from other areas. The isolation rates were 62% and 38%,respectively. Pathogenic analysis showed that a total of 6 strains of bacteria were pathogenic,including 2 strains of Escherichia coli,1 strain of Shigella flexneri,2 strains of Staphylococcus aureus and 1 strain of Streptococcus paris. The six isolates were resistant to clindamycin and sensitive to ciprofloxacin, florfenicol, gentamicin, enrofloxacin and sulfamethoxazole.
