不同虫花棒束孢菌株固体培养粗提物抗病毒活性测定 被引量:1

Antiviral activity of extracts from solid culture of Isaria farinosa strains

摘要 目的对不同虫花棒束孢菌株固体培养物进行有机溶液提取,并测定其抗肠道病毒71型(EV71)、黄热病病毒、日本脑炎病毒和登革病毒的活性。方法选取7个虫花棒束孢菌株,经常规活化、培养后,收集菌丝或培养混合物,采用乙酸乙酯浸泡,提取得到粗提物。利用细胞病变效应(CPE)观察法评价不同粗提物浓度(1.6、8.0、40.0和200.0μg/ml)抗4种供试病毒的活性。结果菌株1705、1785、2138、2140、2142、2185、CBS111113粗提物含量分别为417.1、346.4、380.7、338.0、474.3、458.0、475.9 mg,均为黄褐色油状;菌株2138、2140、2142的粗提物对登革病毒表现出较强的抗病毒活性。其中,菌株2138在200.0μg/ml浓度下对登革病毒有抑制作用,且无明显细胞毒性;菌株2140在40.0μg/ml和200.0μg/ml浓度下对正常细胞无明显毒性,且能够抑制50%的细胞病变;菌株2142在40.0μg/ml时对登革病毒有抑制,且无明显细胞毒性。其他菌株的粗提物对4种供试病毒均无明显抗病毒活性。结论不同来源虫花棒束孢菌株粗提物抗登革病毒活性不同,以菌株2142的抗病毒活性较强。部分菌株粗提物无明显抗病毒活性。 Objectives To extract active components from solid culture of different strains of Isaria farinosa and to determine their antiviral activity against enterovirus EV71, yellow fever virus, Japanese encephalitis virus, and dengue virus. Methods Seven representative strains of I. farinosa were selected, and the mixed fermentation products were extracted after routine activation and culture. The crude extracts were obtained using ethyl acetate. The antiviral activity of different crude extracts(1.6, 8.0, 40.0, and 200.0 μg/ml) against enterovirus EV71, yellow fever virus, Japanese encephalitis virus, and dengue virus was evaluated using a cytopathic effect(CPE) assay. Results A crude extract of the 1705 strain was obtained in a volume of 417.1 mg, a crude extract of the 1785 strain was obtained in a volume of 346.4 mg, a crude extract of the 2138 strain was obtained in a volume of 380.7 mg, a crude extract of the 2 140 strain was obtained in a volume of 338.0 mg, a crude extract of the 2142 strain was obtained in a volume of 474.3 mg, a crude extract of the 2185 strain was obtained in a volume of 458.0 mg, and a crude extract of the CBS111113 strain was obtained in a volume of 475.9 mg. Crude extracts were oily and yellowish-brown. Crude extracts of the 7 strains obtained with ethyl acetate displayed no antiviral activity against enterovirus, yellow fever virus, or Japanese encephalitis virus. Extracts of the strains 2138, 2140, and 2142 had obvious antiviral activity against dengue virus;strain 2140 inhibited dengue virus and had a 50% cytopathic effect at both 40.0 and 200.0 μg/ml, while strain 2138 inhibited the virus at a concentration of 200.0 μg/ml and strain 2142 did so at a concentration of 40.0 μg/ml without any cytopathic effect. Conclusion Crude extracts of different strains of I. farinosa had no activity against enterovirus, yellow fever virus, or Japanese encephalitis virus.
