导致雏鹅痛风新型鹅星状病毒的分离鉴定 被引量:68

Isolation and identification of a new type of astrovirus causing gout in goose

摘要 2017年2—12月,我国山东、安徽、江苏、辽宁、河南及广东等地雏鹅群暴发了一种以痛风为主要特征的传染性疾病。该病主要发生于5~20日龄的雏鹅,病鹅内脏器官及关节腔发生严重的尿酸盐沉积,死亡率最高可达50%,给养鹅业造成了巨大的经济损失。为确定引起该病的病原,本试验对在全国各地采集的143份样品进行了实验室检测及病原的分离鉴定。检测结果显示,禽星状病毒阳性检出率达96.5%(138/143)。对山东平原分离株(SDPY株)ORFlb基因进行了遗传进化分析,结果表明该分离株与已报道的鸡源、火鸡源、鸭源及鹅源星状病毒亲缘关系较远,核苷酸同源性仅为49.5%~67.7%。动物回归试验结果显示,1日龄健康雏鹅于接种SDPY株24h后陆续死亡,剖检表现为心脏、肝脏、肺脏、肾脏等内脏器官及关节腔的尿酸盐沉积与肾脏的出血肿大,与自然发病症状一致。由此确定,导致雏鹅痛风病的病原为新型鹅星状病毒。 A severe infectious disease characterized with gout has affected goslings in Shandong, Anhui,Jiangsu, Liaoning, Henan and Guangdong provinces in China from February to December 2017. The disease mainly infected the goslings aged from 5 to 20 days. The goslings have severe urate deposition in the body cavity and joints with a mortality up to 50% ,which caused serious economic losses to the goose industry. In this study, 143 samples from the nationwide goose farms were tested in laboratory for virus isolation and characterization. PCR results showed that the positive rate of astrovirus is up to 96.5%(138/143). Genomic sequence analysis indicated the SDPY strain is far from chicken,turkey, duck, and geese-origin astroviruses and shared 49.5 %- 67.7% nucleotide identities in ORF1 b gene with them. Pathogencity study showed that the strain is highly virulent when experimentally inoculated into goslings and the main pathological changes concluded renal hemorrhage and urate deposition in the internal organs, such as the heart, liver, lungs and kidneys. The clinical manifestations are consistent with natural incidences. From this, it was determined that the causative agent that caused the gosling visceral gout is a new type of goose astrovirus.
