摘要
目的探讨脑源性神经生长因子(BDNF)在缺糖缺氧复灌条件下对小鼠伏隔核(nucleus accumbens,NAc)脑区胶质纤维酸性蛋白(GFAP)的影响。方法 18只小鼠随机均分为正常组(NAc脑片低糖DMEM培养基培养)、缺糖缺氧复灌组(NAc脑片EBSS培养基培养15min后复灌1h)、缺糖缺氧复灌给药组(NAc脑片EBSS培养基中加入BDNF培养15min后复灌1h)。给予相应的处理后,对GFAP蛋白表达量进行测定。结果正常组GFAP蛋白的相对表达量为0.544±0.053,缺糖缺氧复灌组GFAP蛋白的相对表达量为1.439±0.325,缺糖缺氧复灌给药组GFAP蛋白的相对表达量为0.739±0.121。与正常组相比,缺糖缺氧复灌组GFAP蛋白的相对表达量升高,差异有统计学意义(P<0.01);与缺糖缺氧复灌组相比,缺糖缺氧复灌给药组GFAP的蛋白相对表达量降低,差异有统计学意义(P<0.05);与正常组相比,缺糖缺氧复灌给药组GFAP的蛋白相对表达量升高,差异无统计学意义(P>0.05)。结论 NAc脑区中的胶质纤维酸性蛋白GFAP在缺糖缺氧条件下明显升高,BDNF对星形胶质细胞起抑制作用。
Objective To explore the effect of BDNF on GFAP in NAc slices under the condition of oxygen-glucose depriva- tion. Methods 18 mice were randomly divided into normal group (Culture of low-glucose DMEM Medium in NAc slices), OGD/R group (culture of EBSS medium 15 minutes after reperfusion 1 hour in NAc slices) and OGD/R+BDNF group (culture of EBSS medium adding BDNF 15 minutes after reperfusion 1 hour in NAc slices). After treatment, the expression of GFAP protein was determined. Results The relative expression of GFAP in normal group was 0. 544±0. 053, and the relative expression of GFAP in hypoxia-reperfusion group was 1. 439±0. 325 153, the relative expression of GFAP in the drug-deficient hypoxia-reperfusion group was 0. 739±0. 121. There was a statistically significant difference in the relative expression of GFAP protein in the OGD/R group compared with the normal group (P〈0.01), and there was a statistically significant decreased in the protein relative expression of GFAP in the OGD/R+BDNF group and OGD/R group(P〈0.05). There was no statistically significant difference in GFAP pro- tein relative expression between OGD/R+BDNF group and normal group. Conclusion GFAP in the NAc brain region was signifi- cantly elevated under the condition of oxygen-glucose deprivation, and BDNF inhibits astrocytes.
