3种鲤鲫杂交回交后代染色体核型分析

摘要 采用PHA和秋水仙素体内培养法,取鱼肾细胞,用空气干燥法制备3种回交鲤(鲤鲫杂交♀×鲤♂)、回交镜鲤(鲤鲫杂交♀×镜鲤♂)、回交荷包红鲤(鲤鲫杂交♀×荷包红鲤♂)的染色体。经核型分析,回交鲤三倍体的染色体数为3n=150,核型公式为:3n=60m+24sm+36st+30t,臂比(NF)234;回交镜鲤三倍体的染色体数为3n=150,核型公式为:3n=63m+45sm+12st+30t,臂比(NF)258;回交荷包红鲤三倍体的染色体数为3n=150,核型公式为:3n=66m+36sm+18st+30t,臂比(NF)252。 Metaphase chromosome specimens were prepared from short-term culture of kidney cells with air-drying technique of three kind of backcross fish injected with PHA and colchicine in offspring of three backcrosses of female hybrids [(common carp ♀×crucian carp ♂)backcrossed with male common carp, mirror common carp and red purse carp, respectively]. The chromosome number of the triploid backcross fish with male common carp is shown to be 3n=150, with karyotype of 60 m, 24 sm, 36 st and 30 t chromosomes, and NF=234. The triploid backcross fish with male mirror common carp had chromosome number of 3n=150, and karyotype of63 m, 45 sm, 12 st and 30 t chromosomes, and NF=258. The chromosome number of 3n=150, and the karyotype of 66 m, 36 sm, 18 st and 30 t chromosomes with NF=252 are observed in the triploid of backcross fish with male red purse carp.
