摘要
目的:研究大孔吸附树脂纯化毛冬青总黄酮、总皂苷的工艺条件。方法:以总黄酮、总皂苷的吸附率和转移率为指标,考察上样药液质量浓度、pH、树脂径高比、树脂药材比、洗脱乙醇体积分数等影响因素。结果:毛冬青纯化工艺为上样药液质量浓度0.4 g.mL-1,药液pH 4.5,药材-树脂比1∶8,树脂径高比1∶15,分别用12 BV水洗,10 BV 30%乙醇洗脱,10 BV70%乙醇洗脱得总皂苷。结论:采用大孔吸附树脂可较好地纯化毛冬青总黄酮与总皂苷,纯化后毛冬青总黄酮含量、总皂苷均达50%以上。
Objective： To discuss purification technology conditions of total flavonoids and total saponins from Llex pubescens by macroporous resin.Method：With absorption rate and transfer rate of total flavonoids and total saponins as indexes,investigated concentration of sample liquid,pH,ratio of diameter to height of macroporous resin column,ratio of macroporous resin to crude drug,concentration of eluted ethanol and other factors.Result： Optimum purification conditions were as follows：concentration of sample liquid was 0.4 g·mL-1,pH of sample liquid was 4.5,ratio of crude drug to macroporous resin was 1∶8,ratio of diameter to height of macroporous resin column was 1∶15,total saponins were eluted by 12 BV water,10 BV 30% ethanol and 10 BV 70% ethanol respectively.Conclusion： Macroporous resin could be used to purify total flavonoids and total saponins from L.pubescens,the content of total flavonoid and total saponins reached more than 50%.
