摘要
目的:探讨益肾活血方对嘌呤霉素氨基核苷(PAN)诱导的大鼠肾病综合征的治疗作用及其作用机制。方法:Wistar大鼠随机分为假手术组、PAN模型组、益肾活血方高剂量组(YHF-H)、益肾活血方中剂量组(YHF-M)、益肾活血方低剂量组(YHF-L)、蒙诺治疗组(FP)。采用颈静脉注射结合尾静脉加强注射PAN的方法制作肾病模型。PAN首次注射后第5、10、15、20、25和30天测定24h尿蛋白定量;31d后处死动物,做血清总蛋白、血清白蛋白、总胆固醇、三酰甘油、血清尿素氮、血清肌酐等血生化学检查、肾脏组织光学显微镜和电子显微镜检查以及逆转录聚合酶链式反应(RT-PCR)分析各组大鼠肾脏骨形态蛋白及其基因表达的情况。结果:益肾活血方能有效抑制由PAN所诱导的尿蛋白排泄增高、血清蛋白降低和血脂升高,减轻肾小管间质病变、抑制BMPRⅡ和Smad1在肾组织内的表达。结论:益肾活血方对PAN诱导的大鼠肾病综合征有治疗作用,其机制可能与调节骨形态蛋白表达有关。
A Objective:To investigate the suppressive effect and mechanism of Yishen- Huoxue Fang on nephrotic syndrome induced by puromycin amino nucleoside(PAN). Methods :Wistar rats were randomly divided into six groups- sham operation, PAN - model, PAN - model with high- dosage YHF (YHF - H), PAN - model with median- dosage YHF (YHF- M), PAN - model with low - desage YHF (YHF - L), and PAN - model with Fosinopri (FP). The PAN - model was induced by jugular vein injection combined with tail vein injection of PAN. Quantities of 24 hours urinary protein excretion were examined at days 5th, 10th, 15th, 20th, 25th and 30th. All rats were sacrificed at day 31th for blood biochemistry; kidneys histology and RT- PCR analysis. Results: YHF significantly reduced protein excretion, improved protein content in blood and reduced total cholesterol and triglyceride in blood. YHF also inhibited the interstitial damage induced by PAN. Moreover, YHF was able to reverse BMPRII and Smadl mRNAs abundance caused by PAN. Conclusion: Yishen- Huoxue Fang attenuates PAN- induced kidney injury possibly through bone morphogenetic protein signal transduction pathway.
