摘要
目的探讨应用关节制动的方法制作OA动物模型的可行性及可靠性。方法成年新西兰大白兔28只,左后肢膝关节伸直位石膏管型固定,分别于制动后1、2、4和6周随机处死一只,观察其左后肢膝关节软骨病理改变,了解制动效果,其余动物6周后取左侧膝关节软骨及滑膜标本为实验组,对侧肢体膝关节软骨及滑膜标本为对照组,比较两组软骨及滑膜细胞凋亡情况。结果6周后,无论从大体还是镜下以及细胞凋亡情况,兔左后肢膝关节均呈典型的OA特征。结论膝关节伸直位制动5~6周是一个制作OA动物模型的较为理想的方法。
[Objective] To discuss the feasibility and dependability of the osteoarthritie animal model that get from immobilize the knees of rabbit in full extension using plaster east. [Methods] The left knees of 26 rabbits were immobilized in full extension using plaster east. Specimens of the articular cartilage and synovium obtained from 4 rabbits that had been immobilized for 1, 2, 4, 6 weeks were studied by using histopathologie techniques so that the effect of the immobilization would be observed. At six weeks, the samples of the articular cartilage and synovium obtained from the left knees of 24 rabbits and that from the right knees which was used to be control group. TUNEL assay was used to detect the apoptosis of the articular ehondroeytes and synovial cell. [Results] At six weeks, the typical histopathologic character was seen in the articular cartilage obtained from the left knees of the rabbits. [ Conclusion] It is an ideal model to immobilize the knees of rabbit in full extension using plaster east to get osteoarthritic animal model.
