超声波法和超声波酶法提取灵芝多糖的条件研究 被引量:48

摘要 采用单因子分析和正交试验,以灵芝提取物中多糖提取率和多糖含量为指标,分别对超声波法和超声波协同纤维素酶法中影响多糖提取效果的主要因素进行研究。结果表明,超声波酶法中各因素对灵芝多糖提取率的影响程度依次为酶解温度,pH,酶解时间和酶量。最佳提取条件是酶解温度50℃,pH6.0,酶量2.0%(%对底物),酶解时间20min。该方法与超声波法相比多糖提取率稍低,但提取物中多糖含量相对高出20%以上;此外,该方法还能缩短提取周期,是1种值得借鉴的提取灵芝多糖的方法。 Two methods for extracting polysaccharide from powdered Ganoderma lucidum fruit bodies, ultrasound and ultrasound + cellulase, have been evaluated with respect to polysaccharide extraction rates and polysaccharide levels in crude extracts using monofactorial and orthogonal testing. Factors influencing polysaccharide extraction rates by the ultrasound + cellulase method were, in order of importance, temperature, pH, treatment time and cellulase concentration. The optimal extraction conditions were 50 ℃, pH 6.0, 2. 0% cellulase (relative to the amount of powdered sample) and 20 min extraction time. Polysaccharide extraction rates were slightly lower compared with the ultrasound method but the polysaccharide content in crude extracts exceeded 20% and the extraction time was shorter.
