摘要
目的:建立用毛细管电泳高频电导法测定北豆根中生物碱含量的方法,并比较不同提取方法中各组分含量差异。方法:以融硅毛细管(150μm×60 cm)为分离柱,1 mmol·L^(-1)HAc+2 mmol·L^(-1)NH_4Ac+10 mmol·L^(-1)β-CD+2mmol·L^(-1)SDS 缓冲液为电泳介质,分离电压14.0 kV,用高频电导检测法测定了北豆根药材中粉防己碱和青藤碱的含量。结果:粉防己碱和青藤碱的线性范围分别为12.5~900μg·mL^(-1)和25.0~1.00×10~3μg·mL^(-1),平均回收率分别为96.1%和95.6%。结论:本法可用于北豆根药材中有效成分的定量分离和检测。
Objective: To develop a rapid method for the determination of tetrandrine and sinomenine in Menispermum dauricum DC. by capillary electrophoresis with high frequency conductivity detection. Methods:The buffer contained 1 mmol · L^-1 HAc and 2 mmol · L^-1 NH4Ac with 10 mmol· L^-1β- CD and 2 mmol · L^-1 SDS as additives,and the separation voltage was 14. 0 kV. Results:The linearity was obtained in range of 12. 5 -900 μg · mL^-1 for tetrandrine and 25.0 - 1.00 × 10^3 μg · mL^-1 for sinomenine. The recovery range of these two alkaloids was 93.9% -97.8%. Conclusion:This method is rapid and accurate for the quantitative analysis of alkaloids in Menispermum dauricum DC.
