摘要
目的 :探讨严重创伤应激对糖尿病大鼠血清TNF α、IL 6、NO及氧化应激水平的影响。方法 :以腹腔注射链脲佐菌素 (STZ) ,5 0mg/kg体重制成大鼠糖尿病模型 ,在此基础上以左下肢截肢作为严重创伤应激 ,观察应激后 2h、6h、12h、2 4h及 72h各时间点大鼠血清TNF α、IL 6、NO及氧化应激指标 (SOD、MDA、GSH)水平的变化。结果 :大鼠注射STZ 3d后血糖显著升高。对糖尿病大鼠行截肢后 ,血糖显著升高 (P <0 .0 5 ) ;血清TNF α、IL 6及NO水平均明显升高 ,TNF α及NO水平于应激后 12h达峰值 (P <0 .0 1) ,IL 6于应激后 6h达峰值 (P <0 .0 1) ;血清SOD、GSH水平均明显降低 (P <0 .0 1) ,MDA水平明显升高 (P <0 .0 1) ,于应激后 12h达峰值或谷值。结论 :严重创伤应激可导致糖尿病大鼠TNF α、IL 6及NO水平升高 ,抗氧化应激能力下降 ,进一步加剧糖代谢紊乱。
Objective:To investigate the effect of injury-induced stress on serum TNF-α、IL-6、NO and enzymes of oxidative stress in diabetic rats.Methods: Diabetic rat model was induced by streptozocin and amputation of left lower limb was performed as severe traumatic stress.Serum TNF-α、IL-6、NO and anti-oxidant enzymic activities were evaluated before stress (injury-induced) and 2 h、 6 h、 12 h、 24 h、 72 h after stress respectively.Results: The level of glucose was significantly increased after stress.The level of TNF-α、IL-6、NO were significantly increased,and the peaks of TNF-α、NO were at 12 h after stress,the peak of IL-6 was at 6 h after stress.The level of SOD、GSH were significantly decreased and MDA imcreased after stress.The highest/lowest levels of SOD、GSH、MDA were at 12 h after stress. Conclusion: Injury-induced stress increases the level of TNF-α、IL-6、NO and reduces the antioxygenic ability in diabetes rats,and further impairs glucose regulation.
